Summary: Chromatin Immunoprecipitation (ChIP) is a powerful tool for studying protein/DNA interactions and provides insight into gene regulation, gene expression, chromatin remodeling mechanisms and pathway analysis. However, ChIP can be technically demanding and so far, ChIP studies have been limited mainly to cultured cells and model systems.
Although these samples have played an integral role in the study of
chromatin and epigenetic modifications, discrepancies between ChIP profiles in primary cells and cell lines have been observed. Thus, performing ChIP to identify alterations in histone modification and transcription factor binding profiles directly in human primary cells would be highly valuable.
In this webinar, the current pitfalls and challenges of generating high quality ChIP seq profiles for histone modifications and transcription factors in healthy and diseased biological systems will be discussed. We will review the development and optimization of protocols specifically designed to extract quality chromatin from a range of difficult to ChIP samples. These protocols have been proven capable of overcoming the challenges associated with performing ChIP-seq on primary cells and tissues such as FFPE (Formalin-Fixed, Paraffin-Embedded) samples and PBMCs (Peripheral Blood Mononuclear Cells).
